285 Psoriasis resolution in challenging body areas with ixekizumab: Response trajectories by patient profile clusters over time

To identify patients that benefit most from ixekizumab (IXE), unsupervised machine learning applied to Psoriasis Area and Severity Index (PASI) improvement was used define IXE-response clusters in with psoriasis. Using these identified based on PASI, responses challenging body areas (CBA) as well Static Physicians Global Assessment (sPGA) were assessed through 52 weeks. These analyses included treated on-label doses of IXE pooled two Phase 3 clinical trials, UNCOVER-31 IXORA-S2 .Response trajectories over weeks summarized for sPGA(0,1)/sPGA(0); CBA Nail 0 [NAPSI(0)], Scalp [PSSI(0)], Palmoplantar 100 [PPASI(100)]. Two labelled “Rapid Stable (RAS)” “Moderate” identified. An “Ultimate” sub-group further distinguished within the RAS cluster, an even faster more stable response IXE. In Ultimate clusters, demonstrated marked sustained skin clearance per sPGA (including psoriasis resolution) all compared Moderate cluster. Patients showed a higher earlier PSSI(0), PPASI(100), sPGA(0,1) by Week 8, maintained 52. Rapid complete clearance, including resolution nail, scalp, palmoplantar psoriasis, is achievable patients, especially those clusters. References: 1. Blauvelt, A., et al.
